I have formatted the Word document with page breaks, making it easy to identify responses to the reviewers' comments. It seems that highlighting the article is done in the Word document after the main text. In fact, resubmitting is just like submitting a new paper, with a new number, but including the previous review comments can expedite the process.

I am not a big shot, just an ordinary experimentalist. When replying to the reviewers, there is a section labeled "Response to reviewers" for two reviewers, one Word file for each reviewer's comments. I have divided the responses by page. The first upload is the "Manuscript clear", followed by the "Manuscript modification (highlighted version)". I am not sure if there have been any changes in the submission system for revised manuscripts.
